Abstract
INDONESIA:
Bunga matahari (Helianthus annuus) selain dijadikan sebagai tanaman hias, juga telah dibuktikan memiliki aktivitas farmakologi antiinflamasi. antinyeri hingga antimalaria. Tanaman tersebut diketahui memiliki kandungan flavonoid dan sesquiterpen lakton yang tinggi sehingga diduga memiliki aktivitas antikanker. Tujuan penelitian ini adalah untuk mengetahui aktivitas ekstrak etanol 96% daun bunga matahari terhadap induksi apoptosis, siklus sel, dan ekspresi p53 pada sel kanker serviks HeLa.
Ekstrak daun bunga matahari diperoleh dengan metode ekstraksi maserasi menggunakan bantuan gelombang ultrasonik. Pelarut yang digunakan adalah etanol 96%. Pengujian aktivitas menggunakan nilai IC50 yang diperoleh dengan metode MTT. Analisa induksi apoptosis dan siklus sel menggunakan Flowcytometer dan dilakukan pembacaan dengan program cell quest. pengukuran ekspresi p53 menggunakan Immunocytochemistry dengan pewarna antibodi p53.
Hasil penelitian menunjukkan nilai IC50 Ekstrak daun bunga matahari adalah 377,46 μg/mL; pemberian ekstrak daun dengan konsentrasi IC50 meningkatkan nilai apoptosis sel yaitu sebesar 7,17 % dibanding dengan kontrol (2,9 %) dan meningkatkan kematian sel secara nekrosis (90,44%). Ekstrak daun bunga matahari menyebabkan penurunan jumlah sel pada fase G0-G1 yakni 33,98% dan meningkatkan jumlah sel pada fase S dan G2-M masing-masing secara berturut-turut 17,45% dan 21,28% serta meningkatkan jumlah sel ke arah sub G0-G1(12,87%). Aktivitas ekstrak terhadap p53 menunjukkan skor 0. Pemberian ekstrak daun bunga matahari meningkatkan apoptosis sel, memengaruhi regulasi siklus sel dan tidak menunjukkan peningkatan ekspresi p53.

Sunflower (Helianthus annuus) in addition to being used as an ornamental plant, it also been proved to have anti-inflammatory pharmacological activities. anti-pain up to anti-malaria. The plant had known to high content of flavonoids and sesquiterpen lactones substances so it was predicted to have an anti-cancer activity. The purpose of this research was to knowing The Activity of Ethanol 96% Extract of Sunflower Leaves (Helianthus annuus) through Induction of Apoptosis, Cell Cycle, and the expression of p53 in Cervical Cancer HeLa Cells.
Sunflower extracts were obtained by maceration method using ultrasonic wave. It was using Ethanol 96% as the solvent. The activity test used number of IC50 obtained by MTT assay. The analysis of apoptotic induction and cell cyle used Flowcytometer and read it used cell quest program. p53 expression measurements used immunocytochemistry with p53 colorants antibodies.
The results showed that number of IC50 sunflower leaves extract was 377,46 μg/mL; Treated of sunflower leaves extract IC50 increased the cell apoptosis that was 7,17 % compared to controls (2,9 %) and increased the cell-lethal necrosis (90,44%). The sunflower extracts caused a decrease in the number of cells in the G0-G1 phase i.e. 33,98% and increased the number of cells in the S phase and G2-M each in consecutive 17,45% and 21,28% as well increased the number of cells to sub G0-G1 phase (12,87%). Activity of the extract towards p53 showed a score of 0. The distribution of sunflower leaf extract improved cell apoptosis, influenced cell cycle regulation and did not indicate an increased p53 expression.
