Abstract

INDONESIA:

Gelatin merupakan protein yang diperoleh dari hidrolisis kolagen yang secara alami terdapat pada tulang atau kulit binatang. Karakterisasi profil protein merupakan salah satu aspek penting untuk mengetahui karakteristik gelatin. Penelitian ini bertujuan untuk mengetahui berat molekul protein gelatin dengan metode SDS-PAGE. Pemurnian gelatin menggunakan metode presipitasi amonium sulfat dengan variasi konsentrasi sebesar 40, 50, 60 dan 70%. Hasil analisis SDS-PAGE, tidak menunjukkan perbedaan pada pola pita protein sampel gelatin sebelum dan sesudah dimurnikan. Semua sampel gelatin memiliki 3 pita utama yakni 2 α-chains pada pita protein ~135 kDa (α1 dan α2 chains) dan β-chain pada pita protein ~245 kDa. Kadar protein gelatin sebelum dimurnikan sebesar 71,65 ppm, setelah dimurnikan dengan variasi konsentrasi amonium sulfat yaitu sebesar 61,42; 60,45; 59,89 dan 55,32 ppm.

Gelatin is a protein that is derived from collagen hydrolysis in bone or animal skin naturally. Characterization of protein profiles is one of the important aspects in determining the characteristics of gelatin. The research aims at determining the molecular weight of gelatin protein with SDS-PAGE method. The purification of gelatin used precipitation method of sulfate ammonium with variation of concentration of 40, 50, 60 and 70%. The results of the SDS-PAGE analysis showed no difference in protein band pattern of gelatin samples before and after the purification. All gelatin samples had 3 main bands, namely 2 α-chains on the protein band ~ 135 kDa (α1 and α2 chains) and β-chain on the ~ 245 kDa protein band. Gelatin protein content before purified was 71,65 ppm, after purified by variation of sulfate ammonium concentration, it was 61,42; 60.45; 59.89 and 55.32 ppm.
