Abstract
ABSTRAK
Bakteriosin merupakan senyawa protein yang diproduksi oleh bakteri asam laktat dan memiliki sifat bakterisidal yaitu melawan bakteri patogen yang berbahaya bagi tubuh. Lactobacillus plantarum termasuk bakteri asam laktat yang mampu menghasilkan metabolit sekunder salah satunya yaitu bakteriosin. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi inokulum dan lama fermentasi terhadap produksi bakteriosin oleh Lactobacillus plantarum dan untuk mengetahui pengaruh purifikasi bakteriosin terhadap aktivitas antibakteri bakteriosin.
Produksi bakteriosin dari Lactobacillus plantarum dilakukan menggunakan metode difusi cakram. Produksi bakteriosin yang tinggi identik dengan aktivitas antibakteri yang tinggi. Hasil produksi bakteriosin dipurifikasi menggunakan ammonium sulfat dan dialisis untuk menghilangkan pengaruh dari senyawa lain yang dihasilkan BAL. Kadar protein diuji menggunakan metode biuret dan diukur luas zona hambat sebagai aktivitas antibakteri.
Hasil analisis dengan Two way ANOVA menunjukkan bahwa uji aktivitas antibakteri bakteriosin terhadap Staphylococcus aureus menunjukkan adanya interaksi antara konsentrasi inokulum dan lama fermentasi sedangkan pada Escherichia coli tidak menunjukkan interaksi antara konsentrasi inokulum dan lama fermentasi. Zona hambat yang dihasilkan oleh bakteri Staphylococcus aureus sebesar 5,0 mm dan bakteri Eschericia coli yaitu sebesar 4,2 mm. Hasil purifikasi menujukkan konsentrasi protein sebesar 0,262 mg/ml dengan luas zona hambat sebesar 5, 61 terhadap Staphylococcus aureus dan 6,05 terhadap bakteri Eschericia coli.
ABSTRACT
Bacteriocin is a protein compound produced by lactic acid bacteria (LAB) and has a bactericidal character that is against pathogenic bacteria. Lactobacillus plantarum includes lactic acid bacteria able to produce secondary metabolites like bacteriocin. The aims of this study are to determine the effect of inoculum concentration and fermentation period on bacteriocin production by Lactobacillus plantarum and also to determine the effect of bacteriocin purification on bacteriocin antibacterial activity.
The production of bacteriocin from Lactobacillus plantarum was performed using disc diffusion method. High production of bacteriocin was identically with high antibacterial activity. Bacteriocin extract production was purified using ammonium sulfate and dialysis to remove the effect of other compounds produced by LAB. The protein concentration was tested using the biuret method and the inhibitory zone width measured as an antibacterial activity.
The result of analysis with Two way ANOVA showed that bacteriocin antibacterial activity test against Staphylococcus aureus showed an interaction between inoculum concentration and fermentation period while in Escherichia coli did not show interaction between inoculum concentration and fermentation period. The value of inhibition zone produced by Staphylococcus aureus was 5.0 mm and Eschericia coli was 4.2 mm. Purification results showed that value of protein concentration was 0.262 mg/ml with the value of inhibition zone was 5,61 on Staphylococcus aureus and 6.05 on Escherichia coli.
