Abstract
INDONESIA:
Kurkuminoid merupakan salah satu senyawa dalam rimpang kunyit putih yang dapat dimanfaatkan sebagai obat. Beberapa penelitian menunjukkan bahwa kurkuminoid pada kunyit putih mampu berperan sebagai antikanker. Penelitian ini bertujuan untuk mengetahui aktivitas fraksi etanol (ekstrak kasar) dan isolat kurkuminoid terhadap sel kanker payudara T47D dan sel Vero. Sampel kunyit putih diekstraksi menggunakan metode maserasi dengan pelarut etanol kemudian dipartisi menggunakan n-heksana. Fraksi etanol yang diduga mengandung senyawa kurkuminoid diuji fitokimia dan kromatografi lapis tipis (KLT) dengan eluen terbaik. Fraksi etanol (ekstrak kasar) dan isolat yang diduga senyawa kurkuminoid dilakukan uji aktivitas menggunakan metode MTT (Microculture tetrazolium). Isolat kurkuminoid dilakukan identifikasi gugus fungsi menggunakan spektrofotometer UV-Vis dan FTIR. Berdasarkan pemisahan menggunakan KLT didapatkan 3 dari 5 isolat kurkuminoid yaitu isolat 1, 2 dan 5. Hasil uji aktivitas didapatkan nilai IC50 fraksi etanol (ekstrak kasar) sebesar 153,3g/mL yang ditunjukkan hasil indeks selektivitas sebesar 2,4, sedangkan nilai IC50 isolat kurkuminoid sebesar 6222,2; 173,8; dan 2528,9 g/mL. Hasil uji menunjukkan bahwa fraksi etanol (ekstrak kasar) lebih efektif menghambat pertumbuhan sel kanker payudara T47D dibandingkan isolat kurkuminoid.Analisis UV-Vis menunjukkan bahwa ketiga isolat mempunyai panjang gelombang 357,9-359,0 nm. Hasil FTIR pada ketiga isolat kurkuminoid mengandung gugus fungsi yang khas dalam senyawa kurkuminoid pada bilangan gelombang 3467-3468 cm-1 dan 1632-1638 cm-1. Puncak serapan yang muncul sesuai dengan serapan gugus regangan O-H dari ikatan hidrogen dan C=O pada spektra isolat kurkuminoid.

Curcuminoid is a compound in white turmeric rhizome that can be used as a medicine. Several researches showed that curcuminoid in rhizome is utilized as anticancer. The aim of this research was to determine the activity of ethanol fraction (crude extract) and isolates of curcuminoid on T47D breast cancer and vero cells. White turmeric rizhome was extracted by maceration with 96% ethanol and partitioned with n-hexane. Ethanol fraction that expected curcuminoid compound was anaylized by phytochemical test and thin layer chromatography (TLC). Ethanol fraction (crude extract) and the isolates (expected curcuminoid) was determined anticancer activity by MTT (Microculture tetrazolium) method. Curuminoid isolate was identified by UV-Vis and FTIR. Based on the TLC, there were three of five isolates (isolate 1, 2 and 5) that positive curcuminoid. The anticancer activity result had IC50 value of ethanol fraction (crude extract) about 153,3g/mL with selectivity index 2,4. The IC50 value of the three of isolate (1, 2 and 5) was 6222,2; 173,8; and 2528,9 g/mL. The result of activity study showed that ethanol fraction (crude extract) more effectively inhibited the growth of T47D breast cancer cells than isolates. The UV-Vis data showed that the isolates had a wavelength 357,9 to 359,0 nm. The FTIR spectra showed a typical functional group in the curcuminoid compound at wave numbers 3467-3468 cm-1 and 1632-1638 cm-1. The absorption peaks from analyzed according to the absorption of the O-H stretching group from the hydrogen bond and C = O of carbonyl in the curcuminoid isolate spectra.
