Abstract
INDONESIA:
Tanaman Legundi (Vitex trifolia Linn) memiliki beberapa efek farmakologi antara lain antibakteri, antifungi, insektisida, antialergi, antipiretik maupun antikanker. Daun legundi juga mengandung minyak atsiri dan alkaloid. Senyawa-senyawa tersebut dapat diperoleh melalui kultur kalus. Diduga penambahan konsentrasi 2,4 D dan TDZ dapat memengaruhi pertumbuhan kalus Legundi sehingga produksinya meningkat dan dapat dijadikan acuan konsentrasi ZPT yang paling optimal dalam peningkatan metabolit sekunder. Tujuan penelitian ini untuk mengetahui pengaruh kombinasi 2,4 D dan TDZ terhadap pertumbuhan kalus daun Legundi secara in vitro.
Penelitian ini termasuk jenis penelitian eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 16 perlakuan dan 3 ulangan. Perlakuan dalam penelitian ini ada dua faktor yaitu: konsentrasi 2,4 D meliputi 0 mg/l, 1 mg/l, 2 mg/l, dan 3 mg/l dan konsentrasi TDZ meliputi 0 mg/l, 0,25 mg/l, 0,5 mg/l dan 0,75 mg/l. Data dianalisis dengan Uji ANAVA Two Way α = 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5%.
Hasil dari penelitian menunjukkan adanya pengaruh konsentrasi 2,4 D dan konsentrasi TDZ terhadap induksi kalus daun Legundi. Konsentrasi 2,4 D 3 mg/l berpengaruh nyata terhadap induksi kalus yaitu 11 HST, persentase kalus 75,67% dan berat kalus 0,207 gr. Konsentrasi TDZ 0 mg/l berpengaruh nyata terhadap persentase kalus 67,916% berat kalus sebesar 0,316 gr. Kombinasi 2,4 D 2 mg/l + TDZ 0 mg/l hanya berpengaruh nyata terhadap berat kalus sebesar 0,603 gr dengan tekstur remah dan warna kalus putih. Namun kalus yang memenuhi untuk kalus metabolit ialah pada konsentrasi 2,4 D 1 ml/L + 0,5 TDZ dengan warna kuning bertekstur kompak.

Legundi plants (Vitex trifolia Linn) have several pharmacological effects such as antibacterial, antifungal, insecticide, allergic, antipyretic or anticancer. Legundi leaves also contain essential oils and alkaloids. These compounds can be obtained through callus cultures. It is suspected that the addition of concentration of 2.4 D and TDZ can affect the growth of Legundi callus so that its production increases and can be used as the most optimal reference of ZPT concentration in the improvement of secondary metabolite. The purpose of this research is to know the effect of combination of 2,4 D and TDZ on Legundi leaf callus growth in vitro.
This research was experimental, using a complete randomized design (RAL) with 16 treatments and 3 replications. The treatments in this study were two factors, namely: 2.4 D concentrations include 0 mg /l, 1 mg /l, 2 mg /l, and 3 mg /l and TDZ concentrations include 0 mg / l, 0.25 mg / l , 0.5 mg / l and 0.75 mg / l. Data were analyzed by ANAVA Two Way Test α = 5%. If there are significant differences then continued with Duncan Multiple Range Test (DMRT) with a significant level of 5%.
The results of the study showed that 2.4 D concentration and TDZ concentration effected on Legundi leaf calligence. The concentration of 2,4 D 3 mg / l had significant effect on callus induction ie 11 HST, callus percentage 75,67% and callus weight 0,207 gr. The concentration of TDZ 0 mg / l had a significant effect on callus percentage of 67,916% callus weight of 0.316 gr. The combination of 2,4 D 2 mg / l + TDZ 0 mg / l only had significant effect on callus weight of 0.603 gr with crumbed texture and white callus color. But the callus that fills for metabolite callus is at concentration of 2.4 D 1 ml / L + 0,5 TDZ with a compact textured and yellow color.
