Abstract
INDONESIA:
Jintan hitam (Nigella sativa L.) adalah tanaman yang berasal dari daerah Asia Barat dan Mediterania, yang telah digunakan ribuan tahun sebagai obat dan rempah. di Indonesia jintan hitam masih sepenuhnya diimpor dengan total impor sebanyak 510,333 kg/tahun senilai US$ 364.394. Meningkatnya permintaan Jintan hitam baik pada industri farmasi dan industri rumahan dapat diatasi melalui kultur kalus dengan penambahan 2,4-D dan BAP sehingga produksi budidaya bibit jintan hitam dapat ditingkatkan. Tujuan penelitian ini untuk mengetahui pengaruh 2,4-D dan BAP serta kombinasinya terhadap induksi kalus jintan hitam.
Penelitian ini bersifat eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 25 perlakuan dan 3 ulangan. Terdapat dua faktor dalam penelitian yaitu: konsentrasi 2,4-D meliputi 0 mg/L, 0,5 mg/L, 1 mg/L, 1,5 mg/L dan 2 mg/L dan konsentrasi BAP meliputi 0 mg/L, 0,5 mg/L, 1 mg/L, 1,5 mg/L dan 2 mg/L. Data dianalisis dengan Uji ANAVA Two Way α = 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5% serta uji regresi korelasi.
Hasil penelitian menunjukkan adanya pengaruh pemberian 2,4-D dan BAP. Konsentrasi 2,4-D yang efektif adalah 1,5 mg/L menginduksi hari mucul kalus 16,27 HST, persentase terbentuknya kalus 100% dan berat basah kalus 0,0893 gr. Sedangkan konsentrasi BAP yang efektif adalah 0,5 mg/L menginduksi hari muncul kalus 17,67 HST, persentase terbentuknya kalus 93% dan berat basah kalus 0,08887 gr. Pada hasil kombinasi 2,4-D dan BAP, kombinasi efektif pada konsentrasi 1,5 mg/L 2,4-D + 1 mg/L BAP dengan hari muncul kalus 15,6667, persentase terbentuknya kalus 100%, berat basah kalus 0,0967 gram dengan warna kalus yang dihasilkan adalah putih kekuningan, dan memiliki tekstur remah. Sedangkan hasil pengamatan anatomi menunjukkan sel kalus berukuran kecil dan memiliki inti sel.

Black cumin (Nigella Sativa L.) is a plant originating from the Western Asia and Mediterranean regions, which has been used for thousands of years as medicine and spice. In Indonesia black cumin is still fully imported with total amount of 510,333 kg / year worth US$ 364,394. The growing demand for black cumin both in the pharmaceutical and home industries can be overcome through callus culture with the addition of 2.4-D and BAP so that the production of cumin seed cultivation can be improved. The purpose of this research was to know the effect of 2.4-D and BAP and its combination to the induction of black cumin callus.
This study was experimental, using a completely randomized design (CRD) with 25 treatments and 3 replications. Ther10e were two factors in this study: 2.4-D concentration included 0 mg/L, 0.5 mg/L, 1 mg/L, 1.5 mg/L and 2 mg/L and BAP concentrations included 0 mg/L, 0.5 mg/L, 1 mg/L, 1.5 mg/L and 2 mg/L. Data was analyzed by ANAVA Two Way Test α = 5%. If there were significant differences then Duncan Multiple Range Test (DMRT) was conducted with 5% significant level and also correlation regression test .
Results of research showed the effect of giving 2.4-D and BAP. The effective 2.4-D concentration was 1.5 mg/L induced callus emerged day 16.27 HST, the percentage of callus formation 100 % and wet weight of callus 0.0893 gr. While the effective concentration of BAP was 0.5 mg/L induced callus emerged day 17.67 HST, percentage of callus formation 93% and wet weight of callus 0.08887 gr. In result of combination of 2.4-D and BAP, the effective combination was at concentration of 1.5 mg/L 2.4-D + 1 mg/L BAP with callus emerged day 15,6667, percentage of callus formation 100%, wet weight callus 0.0967 gr with the callus produced was yellowish white in colour, and had a crumb texture. While the observation of anatomy showed small callus cells and had nuclei.
