Abstract
INDONESIA:
Jintan hitam (Nigella sativa L.) merupakan salah satu tanaman yang banyak digunakan dalam pengobatan Islam. Kebutuhan biji jintan hitam terus meningkat, dalam dunia medis thymoquinone digunakan antara lain sebagai; antimikroba, antiparasit, antikanker, antiimflamasi, imunomodulator, antioksidan dan sebagainya. Thymoquinone dapat diperoleh melalui kultur kalus dengan penambahan konsentrasi 2,4-D dan kinetin sehingga produksi metabolit sekundernya meningkat dan dapat dijadikan sebagai protokol konsentrasi ZPT yang paling optimal dalam peningkatan metabolit sekunder. Tujuan penelitian ini untuk mengetahui pengaruh 2,4-D dan kinetin serta kombinasinya terhadap induksi kalus jintan hitam (Nigella sativa L.).
Penelitian ini bersifat eksperimental, menggunakan rancangan acak lengkap (RAL) dengan 35 perlakuan dan 3 ulangan. Perlakuan dalam penelitian ini ada dua faktor yaitu: konsentrasi 2,4-D meliputi 0 mg/L, 0,5 mg/L, 1 mg/L, 1,5 mg/L, 2 mg/L, 2,5 mg/L dan 3 mg/L, konsentrasi kinetin meliputi 0 mg/L, 0,5 mg/L, 1 mg/L, 1,5 mg/L dan 2 mg/L. Data dianalisis dengan Uji ANAVA twoways α = 5%. Apabila terdapat perbedaan signifikan maka dilanjutkan uji Duncan Multiple Range Test (DMRT) dengan taraf signifikan 5% serta uji regresi korelasi.
Hasil dari penelitian menunjukkan adanya pengaruh konsentrasi 2,4-D dan kinetin terhadap induksi kalus jintan hitam. Konsentrasi 2,4-D yang paling efisien untuk semua variabel adalah 1,5 mg/L menginduksi kalus selama 20 HST persentase kalus 63,67% dan berat 0,1452gr. Sedangkan konsentrasi kinetin hanya berpengaruh terhadap berat kalus pada konsentrasi 1 mg/L menginduksi kalus selama 22,29 HST persentase kalus sebesar 54,52% dan berat 0,1190 gr. Kombinasi antara 2,4-D dan kinetin terdapat pengaruh terhadap persentase kalus dan berat kalus, yaitu pada konsentrasi yang efesien 2,4-D 2,5 mg/L + Kinetin 2 mg/L dapat menghasilkan 90% kalus tumbuh dengan berat kalus 0,2028 gr dan warna kalus yang dihasilkan adalah kekuningan dengan tekstur kompak.

Black cumin (Nigella sativa L.) is one of the most widely used plants in the treatment of Islam. The needs for black cumin seeds always increases, especially in the medical world. Thymoquinone is used as; antimicrobial, antiparasitic, anticancer, antiimflamation, immunomodulator, antioxidant, and etc. Thymoquinone can be obtained through callus culture with the addition of 2.4-D concentration and kinetin, so the production of secondary metabolite will increase and it can be used as the most optimal ZPT concentration protocol in the increases of secondary metabolite. The purpose of this study was to investigate the effect of 2,4-D and kinetin and its combination on black cumin callus induction (Nigella sativa L.).
This study was experimental, using a complete randomized design (RAL) with 35 treatments and 3 replications. The treatments in this study consist of two factors, such as: the variation of 2.4-D concentrations that consist of 0 mg / L, 0.5 mg / L, 1 mg / L, 1.5 mg / L, 2 mg / L, 2.5 mg / L, and 3 mg / L, the variation of kinetin concentrations that consist of 0 mg / L, 0.5 mg / L, 1 mg / L, 1.5 mg / L, and 2 mg / L. Data were analyzed by ANAVA test twoways α = 5%. If there are significant differences, it will be continued by Duncan Multiple Range Test (DMRT) with 5% significant level and correlation regression test.
The results of this study showed of the effect of 2,4-D and kinetin on black cumin callus inductions. The most efficient from the variation of 2,4-D concentration for all variables was 1.5 mg / L induced callus for 20 Days After Planting (DAP) callus with the percentage 63.67% and the weight was 0.1452gr. Kinetin concentration only influence on callus weight at concentration 1 mg / L induce callus for 22 Days After Planting (DAP) which the callus percentage equal to 54,52% and its weight 0,1190 gr. A combination of 2,4-D and kinetin has an effect on callus’ percentage and callus’ weight which was at an efficient concentration, such as the combination of 2,4-D 2,5 mg / L + Kinetin 2 mg / L. It can produces 90% of calluses that grow for weight 0, 2028 grams. The result from this study, that the callus color was yellowish with a compact texture.
