Abstract
INDONESIA:
Jagung manis dikenal sebagai komoditas pertanian yang diminati oleh masyarakat. Permintaan yang tinggi tidak diimbangi dengan produksi tanaman jagung manis. Untuk meningkatkan produksi tanaman jagung manis diantaranya dapat dilakukan dengan pemberian pupuk. Penggunaan pupuk anorganik yang menimbulkan banyak dampak negatif perlu dicarikan alternatif pengganti, salah satunya ialah dengan pupuk hayati. Kandidat pupuk hayati dapat berasal dari bakteri endofit. Bakteri endofit yang dapat dijadikan pupuk hayati harus memiliki beberapa kemampuan dalam meningkatkan pertumbuhan tanaman, diantaranya ialah kemampuan dalam menambat N2 di udara dan menghasilkan hormon IAA. Pada penelitian kali ini, dilakukan pengujian potensi bakteri endofit rimpang temulawak dalam menambat N2 di udara dan menghasilkan hormon IAA serta pengaruhnya terhadap pertumbuhan tanaman jagung manis.
Penelitian kali ini menggunakan Rancangan Acak Lengkap (RAL) tunggal. Penelitian dilakukan secara in vitro untuk menguji potensi bakteri endofit dalam menambat N2 di udara dan menghasilkan hormon IAA. Pengujian potensi bakteri endofit dalam meningkatkan pertumbuhan tanaman jagung manis dilakukan secara in vivo. Parameter yang diamati antara lain panjang akar tinggi tanaman, jumlah daun, berat kering tajuk, dan warna daun. Data yang diperoleh dianalisis menggunakan Analysis of Variance (ANOVA) dengan taraf signifikan 5%.
Hasil penelitian menunjukkan isolat Enterobacter cloacae memiliki kemampuan menambat N2 di udara lebih tinggi dibandingkan dengan isolat lain yang ditunjukkan dengan nilai OD sebesar 0,512. Selain itu, Enterobacter cloacae mampu menghasilkan hormon IAA dengan konsentrasi yang paling tinggi yaitu sebesar 0,373 ppm. Pada uji in vivo, Enterobacter cloacae dapat meningkatkan panjang akar sebesar 77%, tinggi tanaman sebesar 44%, jumlah helai daun sebesar 4,5%, dan berat kering tajuk sebesar 91%. Sementara itu, daun tanaman jagung manis yang dilakukan pemberian bakteri endofit memiliki warna yang tidak berbeda jauh dengan kontrol yaitu hijau tua.

Sweet corn is known as the agricultural commodities that are in high demand by the community. Its high demands are not balanced with sweet corn crop production. To increase the production of sweet corn can be done by adding the fertilizer. The uses of inorganic fertilizers that cause a lot of negative impacts need to be looked for alternatives, one of which is biofertilizer. The candidate of biofertilizer can be derived from bacterial endophyte. Bacterial endophyte which can be used as biofertilizer should have some ability in increasing plant growth, among which is the ability in N2 fixation and IAA hormone production. At this time, the research is done to test the potential of bacterial endophyte of wild ginger rhizome in N2 fixation and IAA hormone production as well as its effects on the growth of sweet corn plants.
At this time, research uses Completely Randomized Design (RAL). Research carried out in vitro to test potential bacterial endophyte in N2 fixation and IAA hormone production. To test the potential of bacterial endophyte in increasing the growth of sweet corn plants conducted in in vivo test. The observed parameters are the length of the roots, the height of plant, the amount of leaves, the amount canopy dry weight, and color of the leaves. The data obtained were analyzed using Analysis of Variance (ANOVA) with a significant level of 5%.
The results showed Enterobacter cloacae isolates have the ability to N2 fixation is higher compared to other isolates, indicated by the value of the OD up to 0.512. In addition, Enterobacter cloacae was able to produce the highest concentration of IAA hormone up to 0.373 ppm. On in vivo test, Enterobacter cloacae can increase the length of the root up to 77%, increase height plants up to 44%, the amount of leaves up to 4,5%, increases the amount of canopy dry weight up to91%. Meanwhile, the sweet corn plants leaves which is given by bacterial endophyte has a color that does not vary much with control that is dark green.
