Abstract
INDONESIA:
Buah naga (Hylocereus costaricensis) merupakan tanaman yang berasal dari Meksiko, Amerika Tengah yang telah lama menjadi tanaman budidaya di Indonesia dan buahnya untuk dikonsumsi. Penyakit busuk batang tanaman buah naga memiliki pengaruh pada produktivitas buah naga. Penyakit ini disebabkan oleh fungi patogen yang menginfeksi tanaman sehingga menyebabkan bercak kuning yang semakin lama terjadi pembusukan. Fungi endofit dilaporkan memiliki potensi sebagai agen hayati yang efektif untuk mengendalikan patogen. Penelitian ini bertujuan untuk mengetahui jenis fungi patogen penyebab busuk batang dan potensi fungi endofit Fusarium sp. dan Mucor sp. sebagai antagonis terhadap fungi patogen penyebab penyakit busuk batang tanaman tersebut.
Metode yang digunakan adalah eksplorasi dan eksperimen. Penelitian dilakukan dengan cara mengisolasi dan mengidentifikasi fungi patogen dari kulit buah naga yang diperoleh dari perkebunan buah naga UD. Naga Jaya Makmur Bululawang, Malang, selanjutnya dilakukan identifikasi terhadap fungi patogen yang tumbuh pada media PDA. Kemudian dilakukan uji antagonis fungi endofit Fusarium sp. (F) dan Mucor sp. (M) terhadap fungi patogen menggunakan dual culture secara in vitro.
Hasil penelitian menunjukkan sebanyak 4 isolat fungi patogen berhasil diisolasi dari batang buah naga yang terserang penyakit busuk batang yaitu Monosporium sp. (P1), Neoscytalidium sp. (P2), Fusarium sp.1 (P3), dan Fusarium sp.2 (P4). Hasil uji antagonis menunjukkan persentase penghambatan tertiggi ditunjukkan oleh Mucor sp. dalam menghambat P4 dengan rata-rata persentase hambatan sebesar 59,31%, diikuti MP1 sebesar 55,42%, dan FP3 sebesar 41,52%. Mekanisme penghambatan oleh isolat Fusarium sp. terjadi secara antibiosis yang didifusikan ke dalam media agar sehingga terbentuk zona bening, sedangkan Mucor sp. menghambat fungi patogen dengan mekanisme kompetisi dan mikoparasit.

Dragon fruit is a plant which comes from Mexico, Central America. This plant is being a cultivated plant in Indonesia for a long time and its fruit to be consumed. The fungal infection of dragon fruit plants gives impact to the fruits productivity. This disease is caused by fungi pathogen which later will infect the plants. It causes yellow pockmarked on the plant which will be decaying them. Fungi endophyte is known as a potential biological agent that can control pathogen effectively. The aim of this research is to know the types of fungi pathogen which cause the decaying and the potency of fungi endophyte fusarium sp. And Mucor sp. Which is the antagonist to fungi pathogen as the cause of fungal infection diseases.
The research methods that is used are exploration and experiment. This research is done by isolating and identifying the fungi pathogen which is taken from the skin of the fruits in dragon fruits park UD. Naga Jaya Makmur, Bululawang, Malang. Then, the researcher will identify fungi pathogen that grows in PDA media and do the antagonist test to fungi endophyte Fusarium sp. (F) dan Mucor sp. (M) towards fungi pathogen by using dual culture in vitro.
The result of this research shows that 4 isolat fungi pathogen is isolated from the infected stem of dragon fruits, those are Monosporium sp. (P1), Neoscytalidium sp. (P2), Fusarium sp.1 (P3), dan Fusarium sp.2 (P4). Also, the result of antagonist test shows that the percentage of highest inhibition is Mucor sp. in the way of inhibiting P4 with the average percentage of inhibition 59,31%, which is followed by MP1 55,42%, and FP3 41,52%. The mechanism of inhibition which is done by isolat Fusarium sp. is proceeded in antibiosis diffusion process into media. Therefore, the clear zone will be formed, while for Mucor sp. the inhibition of fungi pathogen will be analysed by using competition and microparasite as the mechanisms.
